Biological Background: CD68 (Macrosialin) Function and Localization

  • Alternative splicing produces multiple CD68 isoforms. CD68 is a heavily glycosylated transmembrane protein belonging to the lysosomal/endosomal-associated membrane glycoprotein (LAMP) family.
  • CD68 plays a key role in phagocytic activities of tissue macrophages, both in intracellular lysosomal metabolism and in extracellular cell-cell and cell-pathogen interactions.
  • It binds to tissue- and organ-specific lectins or selectins, facilitating homing of macrophage subsets to particular tissues.
  • Rapid recirculation of CD68 from endosomes and lysosomes to the plasma membrane may allow macrophages to crawl over selectin-bearing substrates or other cells.
  • Subcellularly, CD68 localizes primarily to endosome and lysosome membranes, but can also be found on the cell membrane.
  • It is highly expressed in blood monocytes and tissue macrophages, and also detected in lymphocytes, fibroblasts, endothelial cells, and many tumor cell lines, where it may aid in dissemination.
  • The protein contains a signal peptide, a transmembrane domain, and several disulfide bonds, and undergoes N-linked glycosylation.

Experimental Guidance and Technical Tips

  • Since CD68 is primarily located in intracellular lysosomal and endosomal compartments, perform appropriate fixation and permeabilization steps before staining for intracellular flow cytometry.
  • Validate antibody performance in relevant macrophage model systems such as THP-1-derived macrophages or primary human monocytes.
  • The fluorophore has excitation/emission similar to Pacific Blue, so ensure your flow cytometer is configured with a violet laser (e.g., 405 nm) and appropriate detection filters (e.g., 450/50 BP).
  • This antibody was raised against the extracellular/lumenal domain (aa 22-319) of human CD68, which may target both surface and intracellular protein pools.
