Biological Background: CD68 Function and Localization

  • Macrosialin (CD68) is a heavily glycosylated transmembrane glycoprotein belonging to the LAMP family, encoded by the CD68 gene in humans.
  • Plays a key role in phagocytic activities of tissue macrophages, including intracellular lysosomal metabolism and extracellular cell-cell/cell-pathogen interactions.
  • Binds to tissue- and organ-specific lectins or selectins, facilitating homing of macrophage subsets to particular sites.
  • Undergoes rapid recirculation from endosomes and lysosomes to the plasma membrane, potentially enabling macrophages to crawl over selectin-bearing substrates or other cells.
  • Localizes to the cell membrane, endosome membrane, and lysosome membrane.
  • Highly expressed in blood monocytes and tissue macrophages; also found in lymphocytes, fibroblasts, endothelial cells, and many tumor cell lines, which may aid in tumor cell adhesion and dissemination.
  • Contains a signal peptide, a transmembrane helix, and multiple disulfide bonds; undergoes alternative splicing.
  • Post-translational modifications include extensive N- and O-linked glycosylation.

Experimental Guidance and Technical Tips

  • For flow cytometry (intracellular staining), permeabilize cells prior to staining to ensure antibody access to intracellular compartments where CD68 is abundant.
  • Validate the antibody in your model system using known positive controls such as human macrophage cell lines (e.g., THP-1) or primary monocytes.
  • Consider using a suitable isotype control and Fc receptor blocking to minimize non-specific binding.
  • Optimize antibody concentration and staining conditions for your specific experimental setup, as CD68 expression levels can vary between cell types.
