Biological Background: CD66b (CEACAM8) Function and Localization
- CD66b (CEACAM8) is a single-pass type I membrane protein and member of the carcinoembryonic antigen (CEA) family, also designated CD67, CGM6, and NCA-95.
- Functions as a calcium-independent cell adhesion molecule, mediating heterophilic interactions with other CEACAM family members such as CEACAM6 (PubMed:11590190, PubMed:2022629, PubMed:8776764). Related references: PMID:11590190, PMID:2022629, PMID:8776764
- Heterophilic adhesion between CEACAM8 molecules occurs specifically on activated neutrophils, contributing to immune cell interaction dynamics (PubMed:8776764). Related references: PMID:8776764
- Localized to the cell membrane and anchored via a glycosylphosphatidylinositol (GPI) moiety; contains immunoglobulin-like domains.
- Expressed predominantly in leukocytes of chronic myeloid leukemia patients and in bone marrow, consistent with a granulocytic lineage marker.
- The mature protein has a predicted molecular weight of ~38 kDa and undergoes N-linked glycosylation and disulfide bond formation.
Experimental Guidance and Technical Tips
- ABflo® 488 conjugate allows direct detection by flow cytometry; no secondary antibody is required. Optimize the antibody dilution (e.g., 0.5–5 µg per 10⁶ cells) using known positive and negative cell lines or primary human neutrophils.
- For surface staining, use live, unfixed cells to preserve epitope integrity; if fixation is necessary, validate that the chosen protocol does not alter CD66b recognition.
- Include appropriate isotype controls and fluorescence-minus-one (FMO) controls to assess background and set gates accurately.
- Not validated for other applications such as IHC, ICC, WB, or ELISA; users should conduct thorough pilot testing if adapting to these platforms.
