- CD63 (also known as granulophysin, LAMP-3, ME491, OMA81H, Tspan-30) is a member of the tetraspanin family, a group of four-transmembrane domain proteins involved in cell adhesion, migration, and signal transduction.
- It functions as a receptor for tissue inhibitor of metalloproteinases 1 (TIMP1) and activates downstream signaling pathways including ITGB1/integrin signaling, leading to AKT, FAK/PTK2, and MAP kinase activation, promoting cell survival, adhesion, spreading, and migration.
- CD63 plays a role in VEGFA signaling by regulating internalization of KDR/VEGFR2, and is essential for normal PMEL luminal domain trafficking, critical for melanocyte development and maturation.
- It is implicated in leukocyte adhesion to endothelial cells through regulation of SELP trafficking, and may be involved in mast cell degranulation in response to specific stimuli.
