Biological Background: P-selectin Function and Localization

  • P-selectin (SELP) is a single-pass transmembrane glycoprotein belonging to the selectin family of adhesion molecules.
  • It functions as a Ca2+-dependent lectin receptor that recognizes sialyl-Lewis X carbohydrates on myeloid cells, mediating tethering and rolling of leukocytes on activated endothelium or platelets.
  • Through its interaction with P-selectin glycoprotein ligand-1 (SELPLG), it initiates leukocyte recruitment during inflammation.
  • It also mediates cell-cell interactions and adhesion via integrins alpha-IIb/beta3 (ITGA2B:ITGB3) and integrin alpha-V/beta-3 (ITGAV:ITGB3) (PubMed:37184585). Related references: PMID:37184585
  • Subcellularly, P-selectin is stored in secretory granules—α-granules of platelets and Weibel-Palade bodies of endothelial cells—and rapidly translocates to the cell surface upon cellular activation by agonists such as thrombin or histamine.
  • The extracellular region comprises a lectin domain, an EGF-like domain, multiple sushi (CCP) repeats, and is modified by N-linked glycosylation and palmitoylation.
  • Primarily expressed in platelets and endothelial cells, its surface expression serves as a marker of platelet and endothelial activation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (aa 42-771) of P-selectin, which includes the lectin and EGF domains, likely providing high specificity for native P-selectin on activated cells.
  • For flow cytometry experiments, consider using human whole blood or isolated platelets stimulated with agonists such as thrombin or histamine to upregulate surface P-selectin expression.
  • Validate antibody performance in the relevant sample system, including appropriate isotype controls and compensation settings for the conjugate.
