Biological Background: L-Selectin Function and Localization
- L-Selectin, encoded by the SELL gene, is also known as CD62L, LAM-1, Leu-8, LECAM1, and lymph node homing receptor, and belongs to the selectin family of cell adhesion molecules.
- It mediates calcium-dependent cell adhesion by binding to glycoprotein ligands on neighboring cells, promoting the initial tethering and rolling of leukocytes on endothelial surfaces – a critical step in lymphocyte homing to peripheral lymph nodes. - L-Selectin is a type I transmembrane glycoprotein located on the cell membrane, featuring an extracellular region with a C-type lectin domain, an EGF-like domain, and short consensus repeat (Sushi) domains.
- The protein is expressed on the surface of most peripheral blood B cells and T lymphocytes, as well as on subsets of natural killer cells, monocytes, and granulocytes.
- Alternative initiation of translation results in two isoforms with predicted molecular weights of approximately 42 kDa and 43 kDa, consistent with the observed doublet in some western blot analyses.
