Biological Background: CD6 Function and Localization
- CD6 is a T-cell differentiation antigen (also known as T12, TP120) that functions as a cell adhesion molecule, mediating cell-cell contacts and regulating T-cell responses through interaction with ALCAM/CD166. Related references: PMID:15048703, PMID:15294938, PMID:16352806
- It acts as a costimulatory molecule, promoting T-cell activation and proliferation, and contributes to signaling cascades downstream of the TCR/CD3 complex. Related references: PMID:24584089, PMID:16914752
- CD6 plays a critical role in the formation and maturation of the immunological synapse, facilitating effective T-cell-APC interactions. Related references: PMID:15294938, PMID:16352806
- It also functions as a calcium-dependent pattern recognition receptor, binding lipopolysaccharide (LPS) from Gram-negative bacteria and lipoteichoic acid from Gram-positive bacteria, leading to MAP kinase activation. Related references: PMID:17601777
- CD6 mediates the activation of inflammatory responses and secretion of pro-inflammatory cytokines upon LPS stimulation. Related references: PMID:17601777
- The protein is localized at the cell membrane and is also found as a soluble secreted form; it is expressed on thymocytes, peripheral blood T-cells, natural killer cells, a subset of B-cells (B-1 cells), and some brain cells. Soluble CD6 is detectable in blood serum. Related references: PMID:15294938, PMID:15048703, PMID:16352806, PMID:17601777, PMID:9013954
- CD6 undergoes alternative splicing, producing multiple isoforms, and is characterized by scavenger receptor cysteine-rich (SRCR) domains, N-linked glycosylation, and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 18-364 of human CD6, encompassing the extracellular domain, suggesting that the antibody recognizes the extracellular region of CD6.
- Conjugated to , a fluorophore with excitation/emission similar to Alexa Fluor 594, suitable for flow cytometry; consider appropriate compensation controls.
- For flow cytometry, titrate the antibody on human PBMCs or CD6-expressing cell lines to determine the optimal staining concentration.
- Include appropriate isotype controls and viability dyes to ensure specificity.
- Validate the antibody in the relevant sample system, such as human T-cells or NK cells, where CD6 is expressed.
