Biological Background: CD59 Function and Localization

  • CD59 is a potent inhibitor of the complement membrane attack complex (MAC), thereby protecting human cells from complement-mediated damage during immune responses.

  • It exerts its inhibitory function by binding to the beta-hairpin structures of the C8 components (C8A and C8B) of the assembling MAC, forming an intermolecular beta-sheet that blocks the incorporation of multiple C9 molecules required for complete pore formation.

  • A soluble form of CD59, present in urine, retains complement-binding activity but exhibits a greatly reduced capacity to inhibit MAC assembly on cell membranes.

  • CD59 is localized at the cell membrane via a glycosylphosphatidylinositol (GPI) anchor and is also secreted into the extracellular environment.
