Biological Background: CD59 Function and Localization
-
CD59 is a potent inhibitor of the complement membrane attack complex (MAC), thereby protecting human cells from complement-mediated damage during immune responses.

PMID:1698710
PMID:2475111 -
It exerts its inhibitory function by binding to the beta-hairpin structures of the C8 components (C8A and C8B) of the assembling MAC, forming an intermolecular beta-sheet that blocks the incorporation of multiple C9 molecules required for complete pore formation.

PMID:11882685
PMID:1698710
PMID:36797260 -
A soluble form of CD59, present in urine, retains complement-binding activity but exhibits a greatly reduced capacity to inhibit MAC assembly on cell membranes.
-
CD59 is localized at the cell membrane via a glycosylphosphatidylinositol (GPI) anchor and is also secreted into the extracellular environment.
-
The protein is glycosylated, contains disulfide bonds, and is known by multiple alternative names such as protectin, MAC-inhibitory protein (MACIP), and membrane inhibitor of reactive lysis (MIRL).
Experimental Guidance and Technical Tips
-
The antibody was raised against a recombinant fragment covering amino acids 26–101 of human CD59. It may recognize both the membrane-bound and soluble forms of the protein; verify specificity in your assay system.
-
For flow cytometry (FC), consider using standard staining protocols with appropriate titration of the antibody to determine optimal concentration for your cell type of interest.
-
CD59 is widely expressed on many human cell types, including erythrocytes and endothelial cells; include positive and negative controls to validate staining.
-
Since CD59 is a GPI-anchored protein, ensure that sample preparation (e.g., cell fixation and permeabilization) is compatible with detection of surface-exposed epitopes.
