Biological Background: CD55 (Complement Decay-Accelerating Factor) Function and Localization
- CD55, also known as decay-accelerating factor (DAF), is a glycophosphatidylinositol (GPI)-anchored membrane protein that belongs to the regulators of complement activation (RCA) family.
- It recognizes C4b and C3b fragments deposited on cell surfaces and interferes with their ability to catalyze the conversion of C2 and factor B, thereby preventing the formation of the C4b2a and C3bBb amplification convertases. Related references: PMID:7525274
- By destabilizing and inhibiting the assembly of C3 and C5 convertases, CD55 protects autologous cells from complement-mediated lysis and tissue damage. Related references: PMID:28657829
- Beyond complement regulation, CD55 serves as a cellular receptor for diverse picornaviruses, including coxsackievirus A21, coxsackieviruses B1, B3, B5, human enterovirus 70 and D68, and echoviruses 6, 7, 11, 12, 20, and 21.
- It is expressed on the plasma membrane of virtually all cells that contact plasma complement proteins, as well as on epithelial cells lining extracellular compartments; soluble forms are also found in body fluids and the extracellular matrix.
- The protein contains multiple N-linked glycosylation sites and four N-terminal sushi (CCP) domains, which are essential for ligand binding; alternative splicing generates GPI-anchored and secreted variants.
- Clinically, mutations in CD55 are linked to CHAPLE syndrome (complement hyperactivation, angiopathic thrombosis, and protein-losing enteropathy), and the molecule carries the Cromer blood group antigens.
- The mature polypeptide has a predicted mass of ~41 kDa, but extensive glycosylation results in a heterogeneous band pattern ranging from 39 kDa to 59 kDa in western blotting.
