Biological Background: CD55 Function and Localization
- CD55, also known as decay-accelerating factor (DAF), is a GPI-anchored membrane protein that protects cells from complement-mediated damage by inhibiting the C3 and C5 convertases. Related references: PMID:7525274 PMID:28657829
- It recognizes C4b and C3b fragments deposited on cell surfaces and interferes with the formation and stability of the C4b2a and C3bBb convertases, preventing amplification of the complement cascade. Related references: PMID:7525274 PMID:28657829
- CD55 is expressed on the plasma membranes of virtually all cell types that are in contact with plasma complement proteins, as well as on epithelial cells lining extracellular compartments; soluble variants are found in body fluids and extracellular matrix.
- Multiple isoforms exist due to alternative splicing, including both membrane-bound and secreted forms, which modulate complement activity locally and systemically.
- The protein carries the Cromer blood group antigens, and mutations in CD55 are associated with CHAPLE syndrome, a disease characterized by early-onset protein-losing enteropathy and thrombotic events.
- CD55 is rich in Sushi (CCP) domains, disulfide bonds, and glycosylation sites, contributing to its structural stability and ligand recognition.
- Beyond complement regulation, CD55 acts as a cellular receptor for several pathogens, including Coxsackievirus A21, Coxsackieviruses B1, B3, and B5, Human enteroviruses 70 and D68, and echoviruses 6, 7, 11, 12, 20, and 21.
- As a GPI-anchored protein, CD55 partitions into lipid rafts, where it cooperates with other membrane-bound complement regulators to maintain immune homeostasis.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular region (amino acids 35–353) of human CD55, suggesting that this antibody may recognize native, surface-expressed CD55—ideal for flow cytometry without permeabilization.
- is a bright, photostable fluorophore (equivalent to Alexa Fluor® 488), suitable for multicolor FC panels; consider titrating the antibody to determine the optimal dilution for your cell type.
- Validate staining specificity by pre-incubating the antibody with recombinant CD55 protein or by using a CD55-negative cell line as a control.
- For accurate measurement of surface CD55, avoid fixation/permeabilization protocols that could strip GPI-anchored proteins from the membrane.
- In combination with other complement-related markers, this antibody can help delineate subsets of immune cells or assess cytoprotective mechanisms on tumor cells.
