Biological Background: CD50/ICAM-3 Function and Localization
-
CD50 (ICAM-3, Intercellular Adhesion Molecule 3) is a heavily glycosylated type I transmembrane protein belonging to the immunoglobulin superfamily, and serves as a key cell adhesion molecule on resting leukocytes.
Related references: PMID:1448173, PMID:1448174, PMID:23775590 -
It functions as a primary ligand for the integrin LFA-1 (ITGAL/ITGB2), mediating intercellular adhesion events essential for immune surveillance, including apoptotic neutrophil phagocytosis by macrophages.
Related references: PMID:1448173, PMID:1448174, PMID:23775590 -
ICAM-3 also interacts with integrin ITGAD/ITGB2, further broadening its role in leukocyte adhesion and trafficking.
Related references: PMID:8777714 -
On dendritic cells (DCs), ICAM-3 binds to the C-type lectin CD209 (DC-SIGN) in a calcium-dependent manner, establishing initial contact with resting T cells and promoting T cell proliferation.
Related references: PMID:10721994 -
CD50 is constitutively expressed exclusively on resting leukocytes (including lymphocytes, monocytes, and neutrophils) but is absent from resting or cytokine-activated endothelial cells.
-
The protein contains five immunoglobulin-like C2-type domains in its extracellular region and undergoes post-translational modifications such as N-linked glycosylation and phosphorylation, which regulate its adhesive functions.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 30-114) corresponds to the N-terminal extracellular domain, which is accessible on live cells; consider staining unfixed leukocytes for optimal recognition in flow cytometry.
- Perform a titration experiment to determine the optimal antibody concentration for your specific cell type and assay conditions, as labeling intensity can vary between leukocyte subsets.
- Include an appropriate isotype control antibody conjugated to to assess nonspecific binding and establish gating thresholds.
- For fixation and permeabilization steps, validate that the epitope recognized by this antibody is preserved, as the extracellular domain may be sensitive to certain treatments.
- This product is validated for flow cytometry; other applications may require additional optimization.
