Biological Background: CD48 Antigen Function and Localization
-
CD48 is a glycosylphosphatidylinositol (GPI)-anchored cell surface glycoprotein belonging to the SLAM family (SLAMF2). It is also known as B-lymphocyte activation marker BLAST-1, BCM1 surface antigen, Leukocyte antigen MEM-102, and Signaling lymphocytic activation molecule 2.
-
CD48 interacts via its N-terminal immunoglobulin domain with cell surface receptors CD244 (2B4) and CD2 to regulate immune cell function and activation. Related references: PMID:12007789 PMID:19494291 PMID:27249817
-
In T-cells, CD48 associates with CD2 and facilitates the recruitment of the Src family kinase LCK and the adaptor LAT to the TCR/CD3 complex, promoting LCK phosphorylation and downstream signaling. Related references: PMID:19494291 PMID:12007789
-
CD48 induces phosphorylation of the ITSMs of CD244, initiating signaling events that lead to formation of the immunological synapse and the directed release of cytolytic granules (perforin and granzymes) by T-lymphocytes and NK cells. Related references: PMID:27249817
-
CD48 is widely expressed on all hematopoietic cells.
-
CD48 is localized to the cell membrane, membrane rafts, and may also be secreted.
-
Post-translational modifications include GPI-anchor attachment, disulfide bond formation, and glycosylation; the unprocessed precursor has a molecular weight of approximately 27.7 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular region of CD48 (amino acids 27-220), likely preserving native conformation; consider using live, non-permeabilized cells for flow cytometry to maintain surface epitope integrity.
- Optimize antibody titration for your specific sample type (e.g., peripheral blood mononuclear cells, cultured lymphocytes, or NK cells) and include appropriate isotype and fluorescence-minus-one controls.
- Since CD48 is widely expressed on hematopoietic cells, validate staining specificity by comparing to known positive and negative cell populations.
- For functional studies, note that CD48 is a GPI-anchored protein; crosslinking may be required for certain activation assays.
