Biological Background: CD48 Function and Localization
- CD48 (BCM1, BLAST-1, SLAMF2) is a glycosylphosphatidylinositol (GPI)-anchored cell surface glycoprotein belonging to the signaling lymphocytic activation molecule (SLAM) family. It is encoded by the CD48 gene and expressed as multiple isoforms due to alternative splicing.
- Through its N-terminal immunoglobulin (Ig) domain, CD48 interacts with cell surface receptors CD2 and CD244/2B4, playing a critical role in regulating immune cell function and activation. Related references: PMID:12007789, PMID:19494291, PMID:27249817
- CD48 participates in T-cell signaling by associating with CD2, thereby recruiting the Src family kinase LCK and the adaptor LAT to the TCR/CD3 complex, which promotes LCK phosphorylation and downstream signal transduction. Related references: PMID:19494291, PMID:12007789
- Upon binding CD244, CD48 triggers phosphorylation of the immunoreceptor tyrosine-based switch motifs (ITSMs) of CD244, leading to the formation of the immunological synapse and directed release of cytolytic granules containing perforin and granzymes by T-lymphocytes and NK-cells. - Subcellularly, CD48 is localized at the cell membrane, enriched in membrane rafts, and also exists in a secreted form following cleavage of the GPI anchor.
- CD48 is widely expressed on all hematopoietic cells, including lymphocytes, monocytes, and granulocytes, consistent with roles in innate and adaptive immunity.
- Key structural features include an extracellular immunoglobulin domain, disulfide bonds, N-linked glycosylation sites, and a C-terminal GPI anchor; it is also subject to alternative splicing and proteolytic shedding.

- The immunogen consists of a recombinant fusion protein covering amino acids 27-220 of human CD48 (NP_001769.2), corresponding to the extracellular domain. This design may favor recognition of both membrane-anchored and soluble CD48 forms. Still, verification in the relevant sample system is recommended.
