Biological Background: CD48 Function and Localization

  • CD48, also known as BLAST-1, SLAMF2, and MEM-102, is a glycosylphosphatidylinositol (GPI)-anchored cell surface glycoprotein encoded by the CD48 gene. It belongs to the SLAM family of receptors.
  • It interacts via its N-terminal immunoglobulin domain with cell surface receptors CD244/2B4 and CD2 to regulate immune cell function and activation. Related references: PMID:12007789 PMID:19494291 PMID:27249817
  • In T-cells, CD48 associates with CD2 to bring the Src kinase LCK and LAT to the TCR/CD3 complex, promoting LCK phosphorylation and signal transduction. Related references: PMID:12007789 PMID:19494291
  • CD48 induces phosphorylation of cytoplasmic immunoreceptor tyrosine-based switch motifs (ITSMs) on CD244, initiating signaling for immunological synapse formation and cytolytic granule release in T-lymphocytes and NK cells. Related references: PMID:27249817
  • Subcellular localization includes the cell membrane, membrane rafts, and also a secreted form.
  • Widely expressed on all hematopoietic cells, facilitating its role in immune surveillance and response.
  • Post-translational modifications include N-glycosylation and a GPI-anchor, which are critical for membrane attachment. The protein contains immunoglobulin-like domains and disulfide bonds.
  • Alternative splicing produces multiple isoforms, potentially modulating function.

Experimental Guidance and Technical Tips

  • The immunogen covers the extracellular domain (aa 27-220), making this antibody suitable for detecting cell surface CD48 in flow cytometry without permeabilization.
  • For intracellular staining, consider using a permeabilization protocol; validate in the relevant cell system.
  • Since CD48 is widely expressed on hematopoietic cells, use appropriate controls such as isotype-matched antibodies and unstained cells.
  • The conjugate allows direct detection; test bright signal on lymphocytes, monocytes, or NK cells.
