Biological Background: CD46 Function and Localization

• Also known as membrane cofactor protein (MCP) and trophoblast leukocyte common antigen (TLX), CD46 is a type I membrane glycoprotein and a member of the regulators of complement activation (RCA) family. Alternative splicing generates multiple isoforms, and mutations are associated with atypical hemolytic uremic syndrome (AHUS2).
• Acts as a cofactor for complement factor I, a serine protease that protects autologous cells from complement-mediated damage by cleaving C3b and C4b deposited on host tissue.
• Functions as a costimulatory factor for T-cells, inducing the differentiation of CD4+ T-cells into regulatory type 1 (Tr1) cells, which suppress immune responses through interleukin-10 secretion, thereby playing a role in preventing autoimmunity.
