Biological Background: CD46 Function and Localization
- Also known as membrane cofactor protein (MCP) and trophoblast leukocyte common antigen (TLX), CD46 is a type I membrane glycoprotein and a member of the regulators of complement activation (RCA) family. Alternative splicing generates multiple isoforms, and mutations are associated with atypical hemolytic uremic syndrome (AHUS2).
- Acts as a cofactor for complement factor I, a serine protease that protects autologous cells from complement-mediated damage by cleaving C3b and C4b deposited on host tissue.
- Functions as a costimulatory factor for T-cells, inducing the differentiation of CD4+ T-cells into regulatory type 1 (Tr1) cells, which suppress immune responses through interleukin-10 secretion, thereby playing a role in preventing autoimmunity.
- May be involved in sperm-oocyte fusion during fertilization.
- Serves as a receptor for several pathogens, including Adenovirus subgroup B2 and Ad3, cultured Measles virus, Herpesvirus 6, and pathogenic bacteria Neisseria and Streptococcus pyogenes, exploiting its immunomodulatory properties to induce an immunosuppressive phenotype in T-cells.
