Biological Background: CD46 Function and Localization

  • Membrane cofactor protein (CD46; MCP), also known as TLX, is a type I transmembrane glycoprotein and a member of the regulators of complement activation (RCA) family. It serves as a cofactor for complement factor I, a serine protease that cleaves C3b and C4b deposited on host tissues, thereby protecting autologous cells from complement-mediated injury.
  • CD46 acts as a costimulatory factor for T-cells, inducing the differentiation of CD4+ T-cells into regulatory T (T-reg) type 1 cells. These T-reg 1 cells suppress immune responses by secreting interleukin-10, contributing to the prevention of autoimmunity.
  • The protein is exploited by a number of viral and bacterial pathogens to modulate immune responses. It serves as a receptor for measles virus, adenovirus subgroup B2 and Ad3, herpesvirus 6/HHV-6, and pathogenic bacteria such as Neisseria and Streptococcus pyogenes.   • CD46 plays a minor role in fiber protein-mediated infection by human adenovirus 55 and adenovirus 14a. Related references: PMID:32581096
  • Subcellularly, CD46 localizes to the inner acrosomal membrane of cytoplasmic secretory vesicles, consistent with its role in fertilization, where it may participate in the fusion of spermatozoa with the oocyte.
  • Expressed on all human cells except erythrocytes, CD46 is implicated in various diseases, including hemolytic uremic syndrome. The protein undergoes alternative splicing and post-translational modifications such as disulfide bond formation and phosphorylation.

  • The conjugate is optimized for flow cytometry (FC); ensure your cytometer is equipped with a suitable laser and filter set for 594 nm excitation and far-red emission detection.

  • Because the immunogen corresponds to the extracellular domain (aa 35–328) of human CD46, the antibody is expected to recognize the native surface antigen; validate reactivity in your cell line or primary cell of interest.
