Biological Background: CD46 Function and Localization

• Membrane cofactor protein (CD46; MCP), also known as TLX, is a type I transmembrane glycoprotein and a member of the regulators of complement activation (RCA) family. It serves as a cofactor for complement factor I, a serine protease that cleaves C3b and C4b deposited on host tissues, thereby protecting autologous cells from complement-mediated injury.
• CD46 acts as a costimulatory factor for T-cells, inducing the differentiation of CD4+ T-cells into regulatory T (T-reg) type 1 cells. These T-reg 1 cells suppress immune responses by secreting interleukin-10, contributing to the prevention of autoimmunity.
• The protein is exploited by a number of viral and bacterial pathogens to modulate immune responses. It serves as a receptor for measles virus, adenovirus subgroup B2 and Ad3, herpesvirus 6/HHV-6, and pathogenic bacteria such as Neisseria and Streptococcus pyogenes.
