- Tumor necrosis factor receptor superfamily member 5, also known as CD40, Bp50, CDW40, and TNFRSF5, is encoded by the CD40 gene in humans.
- Acts as receptor for TNFSF5/CD40LG, transducing TRAF6- and MAP3K8-mediated signals that activate ERK in macrophages and B cells, leading to induction of immunoglobulin secretion. Related references: PMID:31331973
- Localizes primarily to the cell membrane and is also secreted, with expression found in B-cells and in primary carcinomas.
- Contains a signal peptide, a transmembrane helix, and multiple repeats; undergoes post-translational modifications including glycosylation and disulfide bond formation.
