Biological Background: CD4 Function and Localization

  • CD4, also known as T-cell surface glycoprotein CD4, T4, or Leu-3, is a 51 kDa transmembrane glycoprotein and member of the immunoglobulin superfamily.
  • It serves as a co-receptor for the T-cell receptor (TCR) by binding to MHC class II molecules on antigen-presenting cells, thereby enhancing T-cell activation and signaling.
  • Upon MHC class II engagement, CD4 recruits the Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling cascades that promote lymphokine production, adhesion, and T-helper cell differentiation.
  • As the primary receptor for HIV-1, CD4 mediates viral entry and is down-regulated by HIV-1 Vpu. Related references: PMID:12089508 PMID:16331979 PMID:2214026 PMID:17346169
  • CD4 also serves as a receptor for human herpesvirus 7 (HHV-7). Related references: PMID:7909607
  • CD4 is predominantly expressed on the surface of T-helper cells, but is also found on macrophages, dendritic cells, and NK cells, playing roles beyond T-cell help including differentiation and cytokine expression.
  • The protein is localized at the cell membrane and undergoes post-translational modifications such as glycosylation, palmitoylation, and phosphorylation.

Experimental Guidance and Technical Tips

  • The immunogen is a recombinant protein covering amino acids 26–207, which corresponds to the extracellular domain of human CD4. This design targets the surface-exposed region and is suitable for flow cytometry detection of membrane-bound CD4.
  • For flow cytometry, consider titrating the conjugate to determine the optimal staining concentration for your cell type and instrument setup.
  • Validate the antibody in your specific sample system, as expression levels of CD4 can vary among T-helper cells, monocytes, and other immune cell subsets.
  • Ensure appropriate compensation controls are included when using alongside other fluorophores.
