Biological Background: CD38 Function and Localization
- CD38, also known as ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 1, is a multifunctional transmembrane glycoprotein encoded by the CD38 gene (GeneID: 952). Multiple isoforms exist due to alternative splicing.
- It functions as an enzyme that synthesizes cyclic ADP-ribose (cADPR), a second messenger for glucose-induced insulin secretion, and also possesses cADPR hydrolase activity. Related references: PMID:7961800 PMID:8253715
- CD38 also catalyzes the synthesis of the Ca(2+) mobilizer nicotinate-adenine dinucleotide phosphate (NAADP+) from 2'-phospho-cADPR and nicotinic acid. Related references: PMID:16690024
- As a receptor, CD38 binds CD31 on endothelial cells, promoting lymphocyte activation, proliferation, and transendothelial migration (PMID:9551996). In lamina propria T lymphocytes, CD38/CD31 interactions initiate a multistep signaling pathway resulting in activation of LCK and LAT, followed by cytokine release (PMID:11259373). Related references: PMID:9551996 PMID:11259373
- In dendritic cells, CD38-mediated signaling regulates CCL21-driven migration, survival, and Th1-polarizing activity, critical for mature dendritic cell functions. Related references: PMID:16293598
- The protein is localized on the cell surface and cell membrane.
- It is highly expressed in pancreas, liver, kidney, brain, testis, ovary, placenta, malignant lymphoma, and neuroblastoma.
- Post-translational modifications include disulfide bond formation and glycosylation; CD38 is implicated in diabetes mellitus.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 43-300, encompassing the extracellular domain of CD38; this design is suitable for detecting surface-expressed CD38 in flow cytometry.
- For flow cytometry, perform antibody titration using a relevant cell line or primary human lymphocytes to determine optimal staining concentration, and include proper isotype and compensation controls.
- Since CD38 is expressed on various immune cells, validate antibody specificity in your target population (e.g., T cells, B cells, NK cells) using multi-color panels.
- The conjugate is compatible with standard violet laser (405 nm) excitation and far-red emission filters; ensure instrument configuration supports this fluorophore.
