Biological Background: CD38 Function and Localization
- CD38 is a multifunctional transmembrane glycoprotein (ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 1) that synthesizes cyclic ADP-ribose (cADPR), a second messenger for glucose-induced insulin secretion. Related references: PMID:7961800 and PMID:8253715
- CD38 functions as a receptor binding CD31 on endothelial cells, promoting lymphocyte activation, proliferation, and migration across endothelial barriers. Related references: PMID:9551996
- Involved in regulating crucial dendritic cell functions at the mature stage, including CCL21-driven migration, survival, and Th1-polarizing activity. Related references: PMID:16293598
- In lamina propria T lymphocytes, CD38/CD31 cognate interactions initiate a signaling pathway resulting in activation of LCK and LAT, followed by cytokine release. Related references: PMID:11259373
- Preferentially transforms 2'-phospho-cADPR into NAADP(+) at both pH 5.0 and 7.4, and also possesses cADPR hydrolase activity. Related references: PMID:16690024
- Subcellular localization: prominently expressed on the cell surface and cell membrane.
- Tissue specificity: highly expressed in pancreas, liver, kidney, brain, testis, ovary, placenta, malignant lymphoma, and neuroblastoma.
- Post-translational modifications include disulfide bond formation and glycosylation; alternative splicing produces multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 43-300) encompasses the extracellular domain, making this antibody suitable for detecting surface CD38 in live-cell flow cytometry without permeabilization.
- Perform titration and use appropriate isotype controls to optimize flow cytometry staining conditions.
- CD38 expression levels can vary between cell types and disease states; validate the antibody in your specific experimental system.
- Ensure compatibility of the conjugate with your cytometer's laser and filter configuration, and perform compensation as needed.
