Biological Background: CD370/CLEC9A Function and Localization

  • CD370/CLEC9A (C-type lectin domain family 9 member A), also known as DNGR-1, is a type II transmembrane receptor containing a single C-type lectin-like domain.
  • It functions as an endocytic receptor on a small subset of myeloid cells specialized for the uptake and processing of material from dead cells.
  • Recognizes filamentous actin associated with particular actin-binding domains of cytoskeletal proteins, including spectrin, which become exposed upon cell membrane damage.
  • Mediates the cross-presentation of dead-cell associated antigens in a Syk-dependent manner, linking innate sensing of cell death to adaptive immunity.
  • Subcellular localization: predominantly expressed on the cell membrane, anchored via a signal-anchor and a single transmembrane helix.
  • Tissue specificity: highly restricted expression on BDCA3+ dendritic cells and a small subset of CD14+ CD16- monocytes in peripheral blood.
  • Post-translational modifications: predicted glycoprotein with disulfide bonds, consistent with its extracellular C-type lectin domain.
  • The predicted molecular weight of the unmodified protein is approximately 27.3 kDa, with the product detecting a band at 27 kDa (presumably the glycosylated form may migrate slightly differently).

Experimental Guidance and Technical Tips

  • The antibody is directly conjugated to , allowing for immediate flow cytometry without secondary staining steps.
  • Titrate the antibody to determine the optimal concentration for your specific cell type and experimental conditions.
  • Validate performance in human peripheral blood mononuclear cells (PBMCs) or enriched dendritic cell populations.
  • Include appropriate isotype control and viability dye to ensure accurate gating on live, CD370+ cells.
  • For fixation, test compatibility as fluorescence may be affected by common fixatives; consider testing under both live and fixed conditions.
