Biological Background: CD369/Dectin-1 Function and Localization
- Serves as a pattern recognition receptor (PRR) specific for beta-1,3-linked and beta-1,6-linked glucans from pathogenic bacteria and fungi. Related references: PMID:11567029, PMID:12423684
- Mediates phagocytosis of C. albicans conidia. Related references: PMID:17230442
- Activates downstream signaling via SYK recruitment and ITAM motif, leading to NF-kB and MAP kinase p38 pathways, enhancing pro-inflammatory cytokine production in macrophages and dendritic cells (By similarity).
- Induces reactive oxygen species production and T-cell proliferation; binds T-cells in a glycan-independent manner to modulate immune responses (By similarity).
- Alternative splicing generates isoforms with varied molecular weights, reflected in observed protein weights of 5 kDa, 8 kDa, and 13-27 kDa.
- Predominantly localizes to the cell membrane and cytoplasm; highly expressed in peripheral blood leukocytes, dendritic cells, spleen, bone marrow, lung, muscle, stomach, and placenta.
- Contains an extracellular C-type lectin domain, a transmembrane helix, and an ITAM signaling motif; classified as a lectin, immune receptor, glycoprotein, and phosphoprotein.
Experimental Guidance and Technical Tips
- The immunogen (aa 66-247) targets the extracellular domain of CD369/Dectin-1, making it suitable for surface staining in live-cell flow cytometry. Validate staining on human myeloid cell lines (e.g., THP-1, U937) or primary monocytes/dendritic cells.
- conjugate provides direct detection without secondary antibody. Use standard PE-Texas Red filter set (excitation 590 nm, emission 617 nm). Ensure proper compensation when multiplexing with other fluorochromes.
- Multiple protein weights (5-27 kDa) due to glycosylation and isoforms may complicate western blot applications. This antibody is validated for flow cytometry; performance in other assays has not been assessed—experimental validation is recommended if used outside FC.
- Shipment on ice bag ensures product stability; upon receipt, store at -20°C as typical for monoclonal antibodies. Avoid repeated freeze-thaw cycles.
