Biological Background: CRTAM Function and Localization

  • CRTAM (CD355) is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily, characterized by two extracellular Ig-like domains and a cytoplasmic tail involved in signaling.
  • Mediates heterophilic cell-cell adhesion, playing a key role in regulating the activation, differentiation, and tissue retention of various T-cell subsets (By similarity).
  • Interaction with CADM1 promotes natural killer (NK) cell cytotoxicity and interferon-gamma (IFNG) secretion by CD8+ T-cells in vitro, and contributes to NK cell-mediated rejection of CADM1-expressing tumors in vivo. Related references: PMID:15811952
  • Regulates CD8+ T-cell proliferation upon TCR activation and promotes cellular polarization of CD4+ T-cells via SCRIB interaction, impacting cytokine production (IFNG, IL17, IL22) (By similarity).
  • Required for the intestinal retention of intraepithelial CD4+ CD8+ and CD8+ T-cells; interaction with CADM1 on CD103+ dendritic cells facilitates gut-homing and T-cell conversion (By similarity).
  • Expressed predominantly on activated class-I MHC-restricted T-cells (NKT, CD8+), and also in spleen, thymus, small intestine, Purkinje neurons; lower levels in testis, ovary, colon, lung.
