Biological Background: CRTAM Function and Localization
- CRTAM (CD355) is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily, characterized by two extracellular Ig-like domains and a cytoplasmic tail involved in signaling.
- Mediates heterophilic cell-cell adhesion, playing a key role in regulating the activation, differentiation, and tissue retention of various T-cell subsets (By similarity).
- Interaction with CADM1 promotes natural killer (NK) cell cytotoxicity and interferon-gamma (IFNG) secretion by CD8+ T-cells in vitro, and contributes to NK cell-mediated rejection of CADM1-expressing tumors in vivo. Related references: PMID:15811952
- Regulates CD8+ T-cell proliferation upon TCR activation and promotes cellular polarization of CD4+ T-cells via SCRIB interaction, impacting cytokine production (IFNG, IL17, IL22) (By similarity).
- Required for the intestinal retention of intraepithelial CD4+ CD8+ and CD8+ T-cells; interaction with CADM1 on CD103+ dendritic cells facilitates gut-homing and T-cell conversion (By similarity).
- Expressed predominantly on activated class-I MHC-restricted T-cells (NKT, CD8+), and also in spleen, thymus, small intestine, Purkinje neurons; lower levels in testis, ovary, colon, lung.
- Key post-translational modifications include glycosylation and disulfide bond formation. Alternative splicing produces isoforms with predicted molecular weights around 22 kDa and 44 kDa.

- This antibody is directly conjugated to and is validated for flow cytometry (FC). Perform antibody titration to determine the optimal staining concentration for your specific cell type and experimental conditions.
- The immunogen corresponds to the extracellular region (aa 18-287), which may recognize both full-length and soluble isoforms if present. Validate detection of target bands at ~22 kDa and/or 44 kDa in your sample system, noting possible glycosylation or cleavage products.
- Include appropriate isotype controls and fluorescence-minus-one (FMO) controls to ensure specificity. Consider using cells known to express CRTAM (e.g., activated CD8+ T-cells) as positive controls.
- For optimal results, use fresh or properly cryopreserved cells; avoid fixation methods that may mask the epitope.
