Biological Background: CD35 Function and Localization
- CD35, also known as Complement Receptor Type 1 (CR1) or C3b/C4b receptor, is a large transmembrane glycoprotein belonging to the regulators of complement activation (RCA) family, characterized by multiple short consensus repeats (SCRs or Sushi domains).
- It functions as a membrane immune adherence receptor, mediating the capture and clearance of complement-opsonized pathogens and immune complexes by erythrocytes and monocytes/macrophages. Related references: PMID:2963069
- CD35 inhibits spontaneous complement activation by impairing the formation and function of alternative and classical pathway C3/C5 convertases, and acts as a cofactor for factor I-mediated cleavage of C3b and C4b. Related references: PMID:2972794, PMID:8175757
- Upon ligand binding, CD35 contributes to immune regulation by promoting the generation of regulatory T cells from activated helper T cells. Related references: PMID:25742728
- Additionally, CD35 serves as a receptor for Epstein-Barr virus during infection.
- The protein is expressed on the membrane of various cell types, including erythrocytes, a subset of T cells, mature B cells, follicular dendritic cells, monocytes, and granulocytes.
- CD35 features multiple Sushi (SCR) domains, a transmembrane helix, and is subject to N-linked glycosylation; it carries the Knops blood group system antigens.
Experimental Guidance and Technical Tips
- This antibody is directly conjugated to , a bright and photostable fluorophore suitable for direct immunofluorescence detection in flow cytometry. Consider titrating the antibody to determine the optimal concentration for your specific cell type and staining conditions.
- As CD35 is expressed on erythrocytes and various leukocyte subsets, include proper isotype controls and Fc receptor blocking steps to minimize non-specific binding, especially in whole blood or PBMC samples.
- The immunogen corresponds to amino acids 901–1095 within the extracellular domain, which may detect a subset of CD35 isoforms or fragments; confirm reactivity with your target cell population by parallel staining with an established CD35 clone if necessary.
- For intracellular staining, note that CD35 is a transmembrane protein; permeabilization may alter epitope accessibility. Validate in your experimental system.
