Biological Background: CD35 Function and Localization
- Complement receptor type 1 (CR1/CD35) is the C3b/C4b receptor that mediates immune adherence and clearance of complement-opsonized pathogens by erythrocytes and monocytes/macrophages. Related references: PMID:2963069
- CR1 inhibits spontaneous complement activation by impairing the formation and function of the alternative and classical pathway C3/C5 convertases. Related references: PMID:2972794, PMID:8175757
- It serves as a cofactor for factor I-mediated cleavage of C3b to iC3b, C3c, and C3d,g, and of C4b to C4c and C4d, downregulating complement cascades. Related references: PMID:2972794

- CR1 is a single-pass type I membrane protein and is expressed on erythrocytes, subsets of T cells, mature B cells, follicular dendritic cells, monocytes, and granulocytes.
- It contains multiple short consensus repeats (Sushi domains) and is heavily glycosylated with an apparent molecular weight of ~223kDa.
- CD35 also functions as a receptor for Epstein-Barr virus during viral entry.

- Validation of the antibody in the relevant sample system is recommended; optimal working concentrations should be determined empirically by titration.
