Biological Background: CD337/NKp30 Function and Localization
- The protein natural cytotoxicity triggering receptor 3 (NCR3), also known as NKp30 or CD337, is a receptor on NK cells that belongs to the immunoglobulin superfamily. It exists as multiple isoforms due to alternative splicing, resulting in protein weights ranging from ~16 to 21 kDa (observed) with a theoretical molecular weight of ~21.6 kDa.
- NCR3 acts as an activating receptor on natural killer (NK) cells, stimulating cytotoxicity upon binding to extracellular ligands such as BAG6 and NCR3LG1.
- Engagement of NCR3 by BAG6 promotes myeloid dendritic cell (DC) maturation, both by killing immature DCs and by inducing NK cells to release TNF-α and IFN-γ, which in turn drive DC maturation.
- The receptor is selectively expressed by all resting and activated NK cells, with weak expression also detected in the spleen.
- Subcellularly, NCR3 localizes to the cell membrane, consistent with its role as a transmembrane receptor.
- The protein features an immunoglobulin domain, a signal peptide, a transmembrane helix, and disulfide bonds; it is also glycosylated. Keywords from UniProt include alternative splicing, glycoprotein, and immunity.
Experimental Guidance and Technical Tips
- Given that the immunogen corresponds to the extracellular domain (amino acids 19-135), the antibody may detect surface-expressed NCR3 without permeabilization. For intracellular flow cytometry, permeabilization is required; optimize fixation and permeabilization protocols for NK cell lines.
- The observed protein weight range (16–21 kDa) suggests multiple isoforms or post‑translational modifications; validate specificity in your cell system using appropriate controls.
- As NCR3 is selectively expressed on NK cells, include control cell types (e.g., T cells) to confirm specificity in IF/ICC experiments.
- For IF/ICC, test a range of antibody dilutions on paraformaldehyde‑fixed cells; antigen retrieval may improve signal if epitope masking is suspected.
