Biological Background: CD337/NKp30 Function and Localization

  • Natural cytotoxicity triggering receptor 3 (NKp30), also designated CD337, is an activating natural killer receptor encoded by the NCR3 gene (aliases 1C7, LY117). The protein exists in alternatively spliced isoforms.
  • Functions as a cell membrane receptor on natural killer (NK) cells; upon binding of extracellular ligands BAG6 and NCR3LG1, it stimulates NK cell cytotoxicity toward target cells, including tumor cells.
  • Engagement of NCR3 by BAG6 promotes myeloid dendritic cell (DC) maturation by eliminating immature DCs and inducing NK cells to release TNF-α and IFN-γ, which further drive DC maturation.
  • Belongs to the immunoglobulin superfamily and contains an immunoglobulin domain; classified as a glycoprotein with disulfide bonds.
  • Localizes to the cell membrane, consistent with its role as a signaling receptor; the protein includes a signal peptide and a transmembrane helix.
  • Selectively expressed on all resting and activated NK cells, with weak expression detected in the spleen.
  • Recognized as a key receptor in innate immunity and cancer immunosurveillance; its 3D structure has been determined (available in the PDB).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 19–135, which covers the extracellular domain of the mature protein after signal peptide cleavage; this region includes the ligand-binding immunoglobulin domain, supporting its use for detecting cell surface NKp30.
  • For flow cytometry, titrate the antibody to determine the optimal staining concentration. Use appropriate isotype controls and, if combining with other antibodies, ensure compensation settings are correctly adjusted.
  • Although the antibody is validated for FC, consider that NKp30 expression may vary with NK cell activation status; include relevant positive (e.g., IL-2-activated NK cells) and negative controls.
  • As a monoclonal antibody, it offers lot-to-lot consistency; however, each new lot should be validated in the end-user’s specific experimental system.
