  • Myeloid cell surface antigen CD33 (Siglec-3, gp67) is a single-pass type I membrane protein belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family (UniProt P20138).
  • CD33 functions as a sialic acid-binding receptor that preferentially recognizes α2,3- and α2,6-linked sialic acid-bearing glycans (PubMed:7718872). Related references: PMID:7718872
  • The cytoplasmic domain contains two immunoreceptor tyrosine-based inhibitory motifs (ITIMs); upon ligand engagement such as C1q or sialylated glycoproteins, ITIMs are phosphorylated by Src-like kinases (e.g., LCK) (PubMed:10887109, PubMed:28325905).
