Biological Background: CD33 Function and Localization
- CD33 (Siglec-3) is a myeloid cell surface antigen belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family, with alternative names p67 and SIGLEC-3.
- It functions as a cell adhesion and signaling receptor that preferentially binds alpha-2,3- and alpha-2,6-linked sialic acid-bearing glycans, mediating cell-cell interactions and maintaining immune cells in a resting state. Related references: PMID:10611343, PMID:11320212, PMID:15597323, PMID:7718872
- Ligand engagement leads to phosphorylation of ITIM motifs by Src-like kinases (e.g., LCK), recruitment of phosphatases SHP-1 and SHP-2, and subsequent downregulation of signaling pathways, including PI3K-dependent monocyte activation. Related references: PMID:10887109, PMID:28325905, PMID:10206955, PMID:10556798, PMID:15597323
- CD33 is primarily expressed on monocytic/myeloid lineage cells and brain microglia, and localizes to the cell membrane and peroxisomes.
- Post-translational modifications include glycosylation, disulfide bond formation, and phosphorylation; an alternative splicing isoform is reported.
Experimental Guidance and Technical Tips
- For flow cytometry (FC), use standard cell surface staining protocols with live human mononuclear cells or cell lines.
- Titrate the antibody to determine the optimal concentration for your specific sample type and instrument settings.
- Include appropriate Fc receptor blocking steps to minimize non-specific binding to myeloid cells.
- Validate staining specificity using relevant negative controls (e.g., isotype control or CD33-negative cells).
