Biological Background: CD33 Function and Localization
- CD33, also known as Siglec-3 (sialic acid-binding Ig-like lectin 3) or p67, is a transmembrane receptor belonging to the immunoglobulin superfamily and Siglec family of sialic acid-binding lectins. It is primarily expressed on cells of the myeloid lineage.
- CD33 preferentially binds alpha-2,3- and alpha-2,6-linked sialic acid-bearing glycans, mediating cell-cell interactions and maintaining immune cells in a resting state. Ligand engagement triggers phosphorylation of its cytoplasmic immunoreceptor tyrosine-based inhibitory motifs (ITIMs) by Src-like kinases such as LCK, creating docking sites for the protein-tyrosine phosphatases SHP-1 and SHP-2. These phosphatases then dephosphorylate downstream signaling molecules to suppress cellular activation.
