Biological Background: CD31/PECAM1 Function and Localization
- CD31, also known as platelet endothelial cell adhesion molecule (PECAM-1), endoCAM or GPIIA', is a glycoprotein member of the immunoglobulin superfamily that functions as a cell adhesion molecule.
- It is required for leukocyte transendothelial migration (TEM) during inflammation, with phosphorylation of Tyr-690 playing a critical role in TEM and intracellular trafficking.
Related references: PMID:17580308, PMID:19342684 - PECAM-1 mediates both homophilic interactions at endothelial cell junctions and heterophilic binding with CD177 on neutrophils, facilitating cell-cell adhesion and immune cell recruitment.
Related references: PMID:27958302, PMID:17580308 - Homophilic ligation of PECAM-1 regulates phagocytosis: it prevents macrophage engulfment of viable leukocytes while promoting clearance of apoptotic cells.
Related references: PMID:12110892 - The protein modulates bradykinin receptor BDKRB2 activity and downstream ERK1/2 signaling in endothelial cells.
Related references: PMID:18672896 - Subcellularly, PECAM-1 is located on the cell membrane, concentrated at membrane rafts and cell junctions. It is highly expressed on platelets, leukocytes, and endothelial cells, with isoform‑specific tissue distribution (e.g., Delta12 in trachea, Delta14‑15 in lung).

Experimental Guidance and Technical Tips
- The immunogen spans the extracellular domain (aa 28–601), making this antibody suitable for detection of cell surface CD31 by flow cytometry.
- Titrate the antibody for optimal staining of your target cell population (e.g., human PBMCs, endothelial cells, or platelets).
- Account for the conjugate in multicolor panel design; use proper single‑stain controls and consider compensation requirements.
- Include a viability marker to exclude dead cells, which may nonspecifically bind antibody conjugates.
- Consider pre‑incubating cells with Fc receptor blocking reagents to minimize background binding.
