Biological Background: CD317/BST2 Function and Localization

  • Official full name: Bone marrow stromal antigen 2 (BST2), also known as HM1.24 antigen or Tetherin. Encoded by the BST2 gene.
  • Acts as an interferon-induced antiviral restriction factor that directly tethers budding enveloped viruses to the host cell membrane, preventing virus release. Targets diverse viruses including HIV-1, HIV-2, SIV, Ebola, Marburg, Lassa, SARS-CoV, and influenza A. Related references: PMID:18200009 PMID:18342597 PMID:19036818
  • Isoform 1 and isoform 2 have differing antiviral and signaling activities; isoform 2 is resistant to HIV-1 Vpu-mediated degradation. Related references: PMID:23028328 PMID:26172439
  • Subcellular localization: found in trans-Golgi network, cell membrane, membrane rafts, cytoplasm, apical cell membrane, and late endosomes.
  • Tissue specificity: predominantly expressed in liver, lung, heart, and placenta; overexpressed in multiple myeloma cells. High expression in B-cell lineage, T-cells, monocytes, NK cells, and dendritic cells.
  • Post-translational modifications include glycosylation, GPI-anchor, disulfide bonds, and isopeptide bond (ubiquitin-like conjugation).
