Biological Background: CD317/BST2 Function and Localization

  • BST2 (Bone marrow stromal antigen 2), also known as tetherin or HM1.24, is an interferon-inducible antiviral host restriction factor.
  • BST2 directly tethers nascent enveloped virus particles to the infected cell membrane, preventing their release as cell-free virions. This tethering can lead to endocytosis and degradation of trapped virions. Related references: PMID:18200009, PMID:18342597, PMID:19036818
  • It restricts a broad range of mammalian enveloped viruses, including HIV-1, HIV-2, SIV, EIAV, HCV, Ebola, Marburg, influenza A, and SARS-CoV. Related references: PMID:18200009, PMID:18342597, PMID:19179289
  • Beyond viral restriction, BST2 inhibits the cell-surface proteolytic activity of MMP14, leading to decreased activation of MMP15 and suppression of cell growth and migration. Related references: PMID:22065321

  • The immunogen corresponds to the extracellular domain (aa 49-160), making this antibody well-suited for detecting surface BST2 by flow cytometry. Consider titrating the antibody to determine optimal staining concentrations.
  • Since BST2 expression is upregulated by type I interferons, pre-treatment of cells with IFN may enhance detection sensitivity.
  • Due to the GPI-anchor, no permeabilization is required for surface staining; for intracellular detection, optimize fixation and permeabilization protocols.
