Biological Background: NKG2-D type II integral membrane protein Function and Localization

  • Encoded by the KLRK1 gene, NKG2-D (also known as CD314, NKG2D, D12S2489E) is a type II integral membrane protein and a member of the killer cell lectin-like receptor family.
  • Functions as an activating and costimulatory receptor involved in immunosurveillance by binding to stress-inducible MHC class I-related ligands (MICA, MICB, ULBP1-4, RAET1 members) on tumor or virus-infected cells.
  • Upon ligand engagement, triggers calcium influx and downstream signaling cascades that stimulate cytotoxic activity and TNF expression in NK cells.
  • Acts as a costimulatory receptor for the T-cell receptor in CD8+ T cells, amplifying adaptive immune responses and promoting perforin-mediated target cell lysis.
  • Participates in NK cell-mediated bone marrow graft rejection and may regulate NK cell differentiation and survival.
  • Localizes to the cell membrane and is expressed predominantly on NK cells (CD56+CD3-), CD8+ αβ and γδ T cells, and interferon-producing killer dendritic cells (IKDCs).
