Biological Background: FCRL1 Function and Localization
- Encodes a type I transmembrane glycoprotein designated Fc receptor-like protein 1 (FCRL1), also known as FcRH1, hIFGP1, IRTA5, and CD307a.
- Regulates B-cell receptor (BCR) signaling, modulating B-cell activation (PubMed:15479727). Related references: PMID:15479727
- Enhances B-cell activation by recruiting the kinase ABL1, while suppressing ERK activation via GRB2 under homeostatic conditions.
- Belongs to the immunoglobulin receptor superfamily, featuring extracellular immunoglobulin-like domains.
- Localizes to the cell membrane as a single-pass transmembrane protein.
- Primarily expressed by mature B cells in secondary lymphoid tissues (e.g., spleen, lymph node), with higher levels in naïve vs. memory B cells; also detected in heart, kidney, liver, and placenta.
- Subject to alternative splicing, producing isoforms observed at 47kDa, 40kDa, and 22kDa (predicted molecular weight of 47kDa).
- Contains intramolecular disulfide bonds and N-linked glycosylation sites essential for receptor structure.
Experimental Guidance and Technical Tips
- Titrate the conjugate to establish an optimal working dilution for your specific cell type and flow cytometry panel.
- Include appropriate isotype control antibodies to assess and adjust for non-specific binding in single- or multicolor experiments.
- Validate specificity using FCRL1‑negative cell lines or knockout controls, especially given the presence of multiple isoforms.
- For multicolor flow cytometry, account for spectral properties and perform proper compensation.
- The immunogen (extracellular domain aa 17‑303) may recognize all major isoforms; verify expected staining pattern in the relevant biological context.
