Biological Background: CD305/LAIR1 Function and Localization
- CD305/LAIR1 is a leukocyte-associated immunoglobulin-like receptor that functions as an inhibitory receptor on natural killer (NK) cells, B-cells, and T-cells, playing a constitutive negative regulatory role in cytolytic function.
- Upon activation by tyrosine phosphorylation, it recruits and activates the phosphatases PTPN6 and PTPN11, and reduces intracellular calcium increase evoked by B-cell receptor ligation.
- It modulates cytokine production in CD4+ T-cells, down-regulating IL2 and IFNG while inducing transforming growth factor beta; in B-cells, it down-regulates IgG, IgE, IL8, IL10, and TNF secretion.
- Subcellular localization is primarily at the cell membrane, consistent with its role as a transmembrane receptor.
- Tissue specificity: expressed on the majority of peripheral mononuclear cells, including NK cells, T-cells, B-cells, monocytes, and dendritic cells; highly expressed in naive T- and B-cells but absent from germinal center B-cells.
- Protein features include immunoglobulin domains, disulfide bonds, glycosylation, and phosphorylation sites; the mature protein has a molecular weight of approximately 31.5 kDa, though observed weights vary (23/29/31 kDa) due to alternative splicing and glycosylation.

Experimental Guidance and Technical Tips
- This antibody is a direct conjugate with , suitable for flow cytometry (FC) analysis of live cells; consider titrating the antibody to determine the optimal staining concentration for your specific sample.
- The immunogen corresponds to the extracellular domain (amino acids 22–163), so it should recognize the native surface receptor; validate staining specificity with appropriate isotype controls and, if possible, blocking peptides or knockout cell lines.
- CD305/LAIR1 is expressed on various immune cell subsets; include unstimulated controls and consider co-staining with lineage markers to identify cell populations of interest.
- Since the protein exhibits multiple molecular weights due to glycosylation and isoforms, use appropriate protein standards if performing complementary biochemical assays; this product is validated for FC, and performance in other applications should be verified.
