Biological Background: CD305/LAIR1 Function and Localization
- Leukocyte-associated immunoglobulin-like receptor 1 (LAIR1, CD305) is a single-pass type I membrane glycoprotein belonging to the immunoglobulin superfamily. It contains characteristic disulfide bonds, glycosylation sites, an immunoglobulin domain, a signal peptide, and a transmembrane helix.
- Functions as an inhibitory receptor on natural killer (NK) cells, B-cells, and T-cells, exerting a constitutive negative regulatory role on cytolytic function. Activation leads to recruitment of phosphatases PTPN6 and PTPN11 and reduces intracellular calcium evoked by B-cell receptor ligation.
- Modulates cytokine production in CD4+ T-cells: down-regulates IL2 and IFNG while inducing secretion of transforming growth factor beta. In B-cells, it down-regulates IgG, IgE, IL8, IL10, and TNF secretion.
- Inhibits proliferation and induces apoptosis in myeloid leukemia cell lines. It prevents nuclear translocation of NF-kappa-B p65 subunit (RELA) and phosphorylation of I-kappa-B alpha (CHUK) in these cells.
- Subcellular localization is primarily at the cell membrane.
- Expressed on the majority of peripheral mononuclear cells, including NK cells, T-cells, B-cells, monocytes, and dendritic cells. Highly expressed in naïve T-cells and B-cells but absent on germinal center B-cells. Abnormally low expression is observed in naïve B-cells from HIV-1 infected patients and very low expression in NK cells from a patient with chronic active Epstein-Barr virus infection.
- The protein undergoes alternative splicing, tyrosine phosphorylation, and glycosylation. Its calculated molecular weight is approximately 31.5 kDa, but multiple bands (23, 29, 31 kDa) are observed due to post-translational modifications.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 22–163) of human CD305/LAIR1; this may facilitate recognition of native epitopes on live cells. Confirm reactivity in your specific experimental system.
- Recommended for flow cytometry (FC). For other applications (e.g., immunofluorescence, immunohistochemistry), pilot testing is advised.
- Multiple protein bands (23, 29, 31 kDa) are reported; consider this heterogeneity when developing western blot assays and include appropriate controls.
- As an inhibitory receptor with variable expression across immune subsets, include proper isotype controls and gating strategies in multicolor flow panels.
