Biological Background: CD305/LAIR1 Function and Localization
- LAIR1 (CD305) is a leukocyte-associated immunoglobulin-like receptor that belongs to the immunoglobulin superfamily. It is an inhibitory receptor containing two ITIM motifs in its cytoplasmic tail.
- Upon ligand binding, LAIR1 undergoes tyrosine phosphorylation, recruiting the phosphatases PTPN6 and PTPN11 to mediate inhibitory signals. This negatively regulates the cytolytic activity of natural killer (NK) cells, B-cells, and T-cells, and attenuates B-cell receptor-induced calcium mobilization.
- In CD4+ T-cells, LAIR1 modulates cytokine production by down-regulating IL2 and IFNG while promoting secretion of TGF-beta. In B-cells, it reduces IgG and IgE synthesis and suppresses secretion of IL8, IL10, and TNF.
- LAIR1 inhibits proliferation and induces apoptosis in myeloid leukemia cell lines by preventing nuclear translocation of NF-kappa-B p65/RELA and phosphorylation of I-kappa-B alpha/CHUK.
- The receptor also inhibits differentiation of peripheral blood precursors into dendritic cells, highlighting its broad immunoregulatory role.
- Subcellularly, LAIR1 is a single-pass type I transmembrane protein located on the cell membrane, with an extracellular immunoglobulin-like domain and cytoplasmic tail.
- LAIR1 is expressed on most peripheral mononuclear cells, including NK cells, T-cells, B-cells, monocytes, and dendritic cells. Expression is high in naïve T- and B-cells but absent from germinal center B-cells. Notably, expression is reduced in naïve B-cells from HIV-1 patients and very low in NK cells from a patient with chronic active EBV infection.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (residues 22–163), making this antibody suitable for detecting cell surface LAIR1 in non-permeabilized samples.
- For flow cytometry, we recommend titrating the antibody to determine the optimal concentration for your experimental system. Use appropriate isotype controls and include known positive (e.g., human PBMCs) and negative cell populations.
- Since LAIR1 is expressed on multiple immune cell types, combine with lineage markers to analyze specific subsets (e.g., CD3 for T-cells, CD19 for B-cells).
- Signal intensity may vary depending on activation state and disease context; validate staining in relevant sample types.
