Biological Background: CD303/BDCA-2 Function and Localization

  • CD303/BDCA-2 is a transmembrane C-type lectin receptor (CLEC4C) primarily expressed on plasmacytoid dendritic cells, where it mediates antigen capture and presentation to T cells. Related references: PMID:11748283 PMID:21880719 PMID:25995448
  • The receptor specifically binds non-sialylated galactose-terminated biantennary glycans, particularly those containing the Gal(β1-3/4)GlcNAc(β1-2)Man trisaccharide epitope, as well as serum IgG. Related references: PMID:21880719 PMID:25995448
  • Upon ligand binding, CD303/BDCA-2 efficiently targets antigens into processing and peptide-loading compartments for MHC-mediated presentation. Related references: PMID:11748283
  • Activation of CD303/BDCA-2 potently inhibits the induction of IFN-α/β expression in PDCs, suggesting a regulatory role in innate antiviral responses. Related references: PMID:11748283 PMID:21880719
  • The receptor functions as a signaling molecule, capable of activating protein-tyrosine kinases and mobilizing intracellular calcium. Related references: PMID:11748283
  • CD303/BDCA-2 is localized to the cell membrane and is constitutively expressed in immature monocyte-derived dendritic cells, with expression significantly down-regulated upon LPS-induced maturation.
  • The protein is characterized by the presence of a C-type lectin domain, a transmembrane helix, and a signal-anchor sequence; it undergoes alternative splicing and N-linked glycosylation.
  • Predicted molecular weight of the unmodified protein is approximately 25 kDa, with glycosylated forms migrating at around 21-25 kDa.

Experimental Guidance and Technical Tips

  • The immunogen is a recombinant fusion protein covering amino acids 45-213 of human CD303/BDCA-2, which includes the extracellular C-type lectin domain; the resulting antibody is expected to recognize native and denatured forms of the target.
  • This -conjugated monoclonal antibody is validated for flow cytometry (FC) applications; titrate the antibody to determine the optimal working concentration for your specific experimental conditions.
  • For immunophenotyping of plasmacytoid dendritic cells, consider co-staining with other PDC markers and appropriate isotype controls.
  • Ensure proper sample preparation and viability to minimize non-specific binding; use freshly isolated or properly cryopreserved human PBMCs or purified dendritic cells.
  • Avoid prolonged exposure to light and store the antibody at 2-8°C protected from light.
