Biological Background: CD301/CLEC10A Function and Localization

  • CLEC10A (also designated HML, MGL, HML2, CD301, CLECSF13, CLECSF14) is a type II transmembrane C-type lectin receptor encoded by the CLEC10A gene in humans.
  • CLEC10A binds GalNAc-terminated glycans in a Ca²⁺-dependent manner, including Tn antigen (GalNAc-O-Ser/Thr), T antigen (Galβ1-3GalNAc), LacdiNAc (GalNAcβ1-4GlcNAc), core 5/6 O-glycans, and gangliosides GM2/GD2, enabling recognition of tumor-associated carbohydrate antigens. Related references: PMID:15802303 PMID:23507963
  • Acts as a signaling receptor at the interface of APC–T cell interactions: it recognizes Tn antigen on PTPRC/CD45 of effector T cells, downregulates CD45 phosphatase activity, and modulates T cell activation threshold, cytokine production, and proliferation. Related references: PMID:15802303 PMID:16998493 PMID:22213806
  • Functions as an endocytic pattern recognition receptor that internalizes Tn‑terminated immunogens and targets them to endolysosomal compartments for MHC class I and II antigen presentation, contributing to antitumor immunity. Related references: PMID:15802303 PMID:17616966 PMID:17804752
  • Localizes to the cell membrane, early endosome membrane, and lysosome membrane, consistent with its roles in ligand binding, signaling, and internalization.
  • Expressed primarily in myeloid antigen‑presenting cells, including dermal dendritic cells and lymph node APCs, as detected at the protein level.
  • Contains a coiled‑coil domain and disulfide bonds, and is N‑glycosylated. The canonical UniProt sequence predicts a molecular weight of ~35 kDa, while observed variants run at 28, 32, and 35 kDa, likely reflecting alternative splicing or differential glycosylation.
  • Multiple isoforms are produced by alternative splicing, which may account for the observed molecular weight heterogeneity.

Experimental Guidance and Technical Tips

  • This antibody is directly conjugated to , a far‑red fluorescent dye, allowing single‑step staining for flow cytometry without a secondary reagent. Titrate the antibody to determine the optimal staining concentration for your experimental system.
  • The immunogen spans the extracellular domain (amino acids 61–316); therefore, the antibody is suitable for live‑cell surface staining without permeabilization. Confirm binding under native conditions in the relevant cell type.
  • When designing multicolor flow cytometry panels, verify spectral compatibility of with other fluorophores used. Preblocking of Fc receptors on myeloid cells with appropriate blocking reagents may reduce nonspecific binding.
  • Include appropriate isotype controls and fluorescence minus one (FMO) controls to assess background staining and define positive populations.
