Biological Background: CD300F/IREM-1 Function and Localization
- CD300F, also designated CMRF35-like molecule 1, IREM-1, or CLM-1, is an inhibitory receptor belonging to the CD300 family of leukocyte surface proteins. It is expressed as multiple isoforms due to alternative splicing.
- The protein functions as an inhibitory receptor on myeloid cells and mast cells, dampening immune activation signals. Related references: PMID:15549731
- CD300F recognizes externalized phosphatidylserine on apoptotic cells and promotes macrophage-mediated efferocytosis, contributing to immune homeostasis.
- Through binding to ceramide and sphingomyelin, it negatively regulates Fc epsilon RI-dependent mast cell degranulation and allergic inflammation. Related references: PMID:24035150
- CD300F suppresses Toll-like receptor (TLR) signaling by recruitment of SHP-1 and SHP-2 phosphatases, limiting MYD88- and TRIF-dependent pathways. Related references: PMID:22043923
- It may serve as an IL-4 co-receptor, enhancing IL-4/IL-13-induced STAT6 signaling, and also inhibits osteoclastogenesis.
- Subcellularly, CD300F is localized to the cell membrane and is predominantly expressed in monocytes, monocyte‑related cells, spleen, peripheral blood leukocytes, and lung.
- Post‑translational modifications include N‑glycosylation, phosphorylation, and disulfide bond formation, contributing to observed molecular weights of 18, 21, 26, and 32 kDa.
Experimental Guidance and Technical Tips
- The immunogen (aa 20–156) targets the extracellular domain, making this antibody well suited for detecting surface CD300F by flow cytometry.
- As a direct conjugate, it enables single‑step staining without secondary antibodies, minimizing non‑specific binding.
- Because CD300F is selectively expressed on myeloid cells, consider using Fc receptor blocking and appropriate isotype controls to reduce background in monocyte‑ or macrophage‑rich samples.
- Validate expression in your specific cell type; while highest levels are reported in monocytes and monocyte‑derived cells, tissue‑specific expression patterns may vary.
