Biological Background: CD300F/IREM-1 Function and Localization
- Encodes CMRF35-like molecule 1 (CLM-1), a member of the CD300 antigen-like family, also designated CD300LF, IREM-1, IgSF13, NKIR, and LMIR3.
- Acts as an inhibitory receptor on myeloid cells and mast cells; negatively regulates Fc epsilon RI-dependent mast cell activation and allergic responses via binding to ceramide and sphingomyelin (PubMed:15549731, PubMed:24035150). Related references: PMID:15549731 PMID:24035150
- Positively regulates the phagocytosis of apoptotic cells (efferocytosis) by recognizing phosphatidylserine exposed on apoptotic cell surfaces, contributing to immune homeostasis (By similarity).
- Negatively modulates Toll-like receptor (TLR) signaling mediated by MYD88 and TRIF through activation of PTPN6/SHP-1 and PTPN11/SHP-2 (PubMed:22043923). Related references: PMID:22043923
- Localizes primarily to the cell membrane.
- Highly expressed in spleen, peripheral blood leukocytes, monocytes, and lung, with low expression in thymus, heart, brain, placenta, liver, skeletal muscle, kidney, pancreas, prostate, testis, ovary, small intestine, and colon.

- The immunogen spans the extracellular domain (amino acids 20–156), suggesting recognition of the native cell surface receptor; consider using non-permeabilized cell staining protocols for flow cytometry.
- For flow cytometry (FC), a suggested starting concentration is 0.5–5 µg/mL; titrate the antibody to determine optimal concentration for your experimental system and include appropriate isotype controls.
- Validate the antibody in physiologically relevant human sample systems, such as peripheral blood mononuclear cells (PBMCs) or monocyte-derived cell lines, given the high expression in monocytes and leukocytes.
- If testing on tissue, note that CD300F/IREM-1 expression varies by organ; lung and spleen show high expression and may serve as positive control sources.
- Since the target undergoes alternative splicing and post-translational modifications, confirm detection of the expected band(s) in lysates from expressing cells if adapting for western blot; however, this product is validated for flow cytometry only.
