Biological Background: CD300F/IREM-1 Function and Localization
- CD300F/IREM-1 (CMRF35-like molecule 1, also known as CLM-1, IgSF13, NKIR) is a member of the CD300 family of immunoregulatory receptors containing an immunoglobulin (Ig) domain.
- Predominantly localized to the cell membrane; highly expressed in spleen, peripheral blood leukocytes, monocytes, and lung, with weak expression in other tissues; selectively found on monocytes and monocyte-related cells.
- Acts as an inhibitory receptor on myeloid cells and mast cells. Related references: PMID:15549731
- Positively regulates the phagocytosis of apoptotic cells (efferocytosis) via phosphatidylserine (PS) recognition; binds PS exposed on apoptotic cells, playing a critical role in immune homeostasis by promoting macrophage-mediated efferocytosis and inhibiting dendritic cell-mediated efferocytosis.
- Negatively regulates FcεRI-dependent mast cell activation and allergic responses by binding ceramide and sphingomyelin as ligands. Related references: PMID:24035150
- May act as a coreceptor for interleukin 4 (IL-4), associating with IL-4 receptor α and augmenting IL-4- and IL-13-induced signaling.
- Negatively regulates Toll-like receptor (TLR) signaling via MYD88 and TRIF by recruiting and activating tyrosine phosphatases SHP-1 (PTPN6) and SHP-2 (PTPN11). Related references: PMID:22043923
- Inhibits osteoclast formation and induces macrophage cell death upon engagement.
- Contains disulfide bonds, is a glycoprotein and phosphoprotein; alternative splicing generates multiple isoforms with observed protein weights of 18 kDa, 21 kDa, 26 kDa, and 32 kDa, reflecting glycosylation and isoform variation.
Experimental Guidance and Technical Tips
- The antibody is supplied as an conjugate, ready for flow cytometry without a secondary detection step; consider titration to optimize staining of human myeloid cell lines or primary monocytes.
- The immunogen corresponds to the extracellular domain (amino acids 20–156), making it suitable for detecting native cell surface CD300F.
- Include appropriate isotype controls and Fc receptor blocking to reduce non-specific binding, especially in monocyte/macrophage samples.
- Since CD300F expression may vary with cell activation or disease state, validate the antibody in the relevant biological context and consider co‑staining with lineage markers.
