Biological Background: CD300a/c Function and Localization

  • CD300a (CMRF35-like molecule 8, also known as IRp60, CMRF35-H9, or IgSF12) is an inhibitory receptor of the CD300 family, which includes both inhibitory and activating members such as CD300c. The antibody recognizes both CD300a and CD300c.
  • It negatively regulates natural killer (NK) cell cytolytic activity and mast cell degranulation, contributing to immune homeostasis. Related references: PMID:10746781 PMID:16339535 PMID:9701027
  • CD300a inhibits Toll-like receptor (TLR) signaling mediated by MYD88 through activation of phosphatase PTPN6, but not the TRIF pathway. Related references: PMID:22043923
  • It is a single-pass type I membrane protein expressed on the cell surface of various immune cells, including NK cells, T-cell subsets, B-cells, dendritic cells, mast cells, granulocytes, and monocytes.
  • CD300a is a glycoprotein with disulfide bonds and phosphorylation sites; alternative splicing produces multiple isoforms.
  • Upon phosphorylation, the cytoplasmic tail of CD300a recruits phosphatases such as PTPN6, which are critical for its inhibitory signaling function.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (amino acids 18–178) of human CD300a, suggesting that the antibody likely recognizes the native surface antigen on live cells, making it suitable for flow cytometry.
  • Since the antibody is pre-conjugated with , it is ready-to-use without secondary staining; titrate carefully to determine the optimal concentration for your specific cell system.
  • Validate antibody staining on human peripheral blood mononuclear cells (PBMCs) or cell lines known to express CD300a/c. Consider using appropriate isotype controls and Fc receptor blocking reagents to reduce nonspecific binding.
  • Be aware that the antibody may cross-react with both inhibitory CD300a and activating CD300c receptors; interpret functional data accordingly and verify target specificity if needed.
