Biological Background: CD273/PD-L2 Function and Localization
- PD-L2 (Programmed cell death 1 ligand 2), also known as B7-DC and CD273, is a member of the B7 family of immune regulatory proteins.
- It plays a critical role in induction and maintenance of immune tolerance to self.
- PD-L2 functions as a ligand for the inhibitory receptor PD-1 (PDCD1), inhibiting T-cell proliferation by blocking cell cycle progression and cytokine production. Related references: PMID:11224527 PMID:12538684
- The protein localizes to the cell membrane and endomembrane system, and can also be secreted.
- Tissue expression is broad, with high levels in heart, placenta, pancreas, lung, and liver, and lower levels in spleen, lymph nodes, and thymus.
- As a glycoprotein with immunoglobulin-like domains, PD-L2 contains disulfide bonds and a transmembrane helix, characteristic of immune checkpoint molecules.

- The immunogen corresponds to the extracellular domain (amino acids 20-219) of human PD-L2, which may allow the antibody to detect native surface-expressed PD-L2 in flow cytometry. Validate reactivity in the relevant human cell or tissue sample system.
