Biological Background: CD271/NGFR Function and Localization
- CD271/NGFR is a member of the tumor necrosis factor receptor superfamily, also known as p75NTR, that functions as a low-affinity receptor for neurotrophins including NGF, BDNF, NTF3, and NTF4. It can form a heterodimeric receptor with SORCS2 that binds precursor neurotrophins with high affinity while showing lower affinity for mature NGF and BDNF. Related references: PMID:24908487
- The receptor plays a critical role in the differentiation and survival of specific neuronal populations during development, and can mediate both cell survival and cell death of neural cells.
- CD271/NGFR is involved in the inactivation of the small GTPase RHOA, influencing cytoskeletal dynamics and cell morphology. Related references: PMID:26646181
- It contributes to insulin-regulated glucose uptake by regulating the translocation of GLUT4 to the cell surface in adipocytes and skeletal muscle cells, likely through modulation of RAB31 activity.
- Necessary for the circadian oscillation of clock genes (BMAL1, PER1, PER2, NR1D1) in the suprachiasmatic nucleus and liver, and of genes involved in glucose and lipid metabolism in the liver. Related references: PMID:23785138
- Together with BFAR, CD271/NGFR negatively regulates NF-kappa-B and JNK signaling pathways, modulating inflammatory and apoptotic responses. Related references: PMID:22566094
- Subcellular localization includes the cell membrane, cytoplasm, perikaryon, and cell projections such as growth cones and dendritic spines, consistent with its role in neuronal signaling and synaptic function.
- The protein undergoes post-translational modifications including glycosylation, phosphorylation, and disulfide bond formation, which may regulate its ligand binding and signaling activities.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 29-250) of human CD271/NGFR, suggesting the antibody is likely to recognize the native conformation of the receptor on the cell surface, making it suitable for flow cytometry.
- For FC applications, use appropriate isotype controls and titrate the antibody to determine the optimal concentration for your specific cell type.
- Since the target is expressed on neuronal cells and certain immune cell subsets, consider validating the antibody in relevant primary cells or cell lines (e.g., SH-SY5Y, PC12, or peripheral blood mononuclear cells).
- The detected protein weight varies (35-45 kDa), possibly due to differential glycosylation or the existence of splice variants; this may affect detection in denaturing applications like Western blot.
- If intracellular staining is required, optimize fixation and permeabilization protocols as the epitope may be sensitive to harsh treatment.
