Biological Background: CD27 Function and Localization
- CD27, also known as TNFRSF7 or T14, is a member of the tumor necrosis factor receptor superfamily and functions as a costimulatory immune-checkpoint receptor expressed on T-cells, NK-cells, and B-cells.
- The receptor binds to its ligand CD70 (CD27L) on B-cells, triggering a signaling pathway that mediates antigen-specific T-cell activation and expansion, thereby providing immune surveillance of B-cells.
Related references: PMID:28011863 - Upon CD70 ligation, CD27 activates the TRAF2-PTPN6 axis, which inhibits LCK phosphorylation to drive phenotypic and transcriptional adaptations necessary for T-cell memory formation.
Related references: PMID:38354704 - In early progenitor cells, CD27 activation by CD70 delivers a negative feedback signal that modulates leukocyte differentiation during hematopoiesis.
- CD27 negatively regulates the function of Th2 lymphocytes in adipose tissue.
- The protein is a single-pass type I membrane receptor localized at the cell surface. It contains disulfide bonds, is N-glycosylated, and is regulated by phosphorylation.
- CD27 is predominantly expressed on most T-lymphocytes and is also found on NK-cells and B-cells. Genetic variations in CD27 are associated with lymphoproliferative disorders.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 21-192) of human CD27, ensuring the antibody recognizes the native cell-surface receptor.
- For flow cytometry, titrate the antibody to determine the optimal concentration for your specific cell type and staining conditions. Include appropriate isotype controls and consider using Fc receptor blocking reagents to minimize nonspecific binding.
- CD27 can be shed from the cell surface upon activation; therefore, timing of sample collection after stimulation may impact detection levels.
- This -conjugated antibody is suitable for multicolor flow cytometry panels; confirm compatibility with other fluorophores in your instrument configuration.
