Biological Background: CD26/DPP4 Function and Localization
- CD26/DPP4 is a multifunctional type II transmembrane glycoprotein belonging to the serine protease family, also known as dipeptidyl peptidase 4, ADABP, and T-cell activation antigen CD26.
- It acts as a costimulatory receptor critically involved in T-cell receptor (TCR)-mediated T-cell activation by binding ADA, CAV1, IGF2R, and PTPRC, thereby positively regulating T-cell coactivation and proliferation.
Related references:
PMID:10900005 PMID:10951221 PMID:11772392 - In concert with fibroblast activation protein (FAP), DPP4 participates in pericellular proteolysis of the extracellular matrix (ECM), facilitating endothelial cell migration and invasion.
Related references:
PMID:10593948 PMID:16651416 - Its serine exopeptidase activity removes N-terminal dipeptides from polypeptides with a penultimate proline, thereby regulating circulating chemokines, growth factors, neuropeptides, and hormones such as brain natriuretic peptide 32.
Related references:
PMID:10570924 PMID:16254193 - DPP4 is localized at the cell membrane, particularly apical membranes, as well as at cell junctions, membrane rafts, invadopodia, and lamellipodia; it is also secreted.
- It is specifically expressed in lymphatic vessels of the skin, small intestine, esophagus, ovary, breast, and prostate, but not in blood vessels, and is found in crypt and villous cells of the small intestine.
- Post-translational modifications include disulfide bond formation and glycosylation, which are essential for its stability and function.
- Additionally, DPP4 serves as the receptor for human coronavirus MERS-CoV, mediating viral entry.
Experimental Guidance and Technical Tips
- The conjugate enables direct detection of CD26/DPP4 without secondary reagents, minimizing nonspecific binding in flow cytometry.
- This antibody, raised against a recombinant protein spanning amino acids 29–766 (the extracellular domain), is expected to recognize native cell surface DPP4; however, titration and appropriate isotype controls should be employed to optimize staining.
- For activation studies or enzymatic activity assays, ensure that fixation/permeabilization protocols do not compromise the epitope or fluorophore.
- If exploring other applications (e.g., immunohistochemistry or Western blotting), validate performance in the relevant sample system, as primary validation is limited to flow cytometry.
