Biological Background: CD26/DPP4 Function and Localization
- Functions as a cell surface glycoprotein receptor that provides costimulatory signals essential for TCR-mediated T-cell activation, acting as a positive regulator by interacting with ADA, CAV1, IGF2R, and PTPRC; interaction with CAV1 and CARD11 promotes T-cell proliferation and NF-κB activation. Related references: PMID:10900005, PMID:10951221, PMID:11772392, PMID:17287217
- Exhibits serine exopeptidase activity, sequentially removing N-terminal dipeptides from polypeptides with unsubstituted N-termini and penultimate proline, thereby regulating various physiological processes by processing chemokines, growth factors, neuropeptides, and peptide hormones. Related references: PMID:10593948, PMID:10570924, PMID:16254193
- In association with FAP, mediates pericellular proteolysis of the extracellular matrix (ECM), facilitating endothelial cell migration and invasion. Related references: PMID:10593948, PMID:16651416
- Involved in cell adhesion and migration, promoting lymphocyte-epithelial cell adhesion via ADA interaction and enhancing lymphatic endothelial cell adhesion and tube formation. Related references: PMID:11772392, PMID:18708048
- Acts as a receptor for human coronavirus MERS-CoV-2.
- Localized to the cell membrane, including apical membrane, invadopodium, lamellipodium, and membrane rafts; also secreted.
- Expressed specifically in lymphatic vessels of the skin, small intestine, esophagus, ovary, breast, and prostate, but not in blood vessels; present in crypt and villous cells of the small intestine, with very low expression in the colon.
- Classified under UniProt keywords such as Aminopeptidase, Cell adhesion, Cell junction, Cell membrane, Host-virus interaction, Hydrolase, Protease, Receptor, Secreted, Serine protease, and Transmembrane; glycoprotein with disulfide bonds.
Experimental Guidance and Technical Tips
- The immunogen encompasses the entire extracellular domain (amino acids 29–766), so the antibody is expected to recognize the native conformation of CD26/DPP4, making it suitable for flow cytometry.
- For FC, titrate the antibody to determine the optimal working concentration, and include appropriate isotype and buffer controls to minimize background.
- Validate reactivity in human samples; cross-reactivity with other species has not been reported and should be tested if required.
