Biological Background: CD263/TRAIL-R3 Function and Localization
- Encodes tumor necrosis factor receptor superfamily member 10C (TNFRSF10C), also known as decoy receptor 1 (DcR1), TRAIL receptor 3 (TRAIL-R3), or lymphocyte inhibitor of TRAIL. It is a GPI-anchored membrane protein that lacks an intracellular death domain.
- Acts as a decoy receptor for the cytotoxic ligand TRAIL (TNFSF10). By competitively binding TRAIL, it inhibits apoptosis induction through functional death receptors TNFRSF10A (DR4) and TNFRSF10B (DR5), thereby protecting cells from TRAIL-induced apoptosis.
- Localized to the cell membrane via a glycosylphosphatidylinositol (GPI) anchor, as indicated by its lipoprotein and GPI-anchor keywords.
- Higher expression observed in normal tissues compared to tumor cell lines, with prominent presence in peripheral blood lymphocytes, spleen, skeletal muscle, placenta, lung, and heart.
- Post-translational modifications include disulfide bond formation and N-linked glycosylation (glycoprotein keyword). The signal peptide is cleaved to yield a mature protein of approximately 27 kDa.
- Member of the TNF receptor superfamily, characterized by cysteine-rich repeats in the extracellular domain.
